The purpose of this book is to familiarize the membrane biologist with the general theory and experimental approaches for membrane protein crystallization. The book begins with a comprehensive explanation of general theory and experimental approaches of crystallization followed by a detailed description of two of the more recent and successful membrane protein expression systems (eukaryotic cell‐free expression and a novel photosynthetic bacterial expression system). Traditional expression systems for both prokaryotic and eukaryotic membrane proteins are summarized in subsequent chapters that address a variety of membrane protein crystallization techniques. Structural themes and biological implications resulting from X‐ray crystallographic structures for specific membrane protein classes are also reviewed.
